NDUFB9 (HGNC:7704): (NADH:ubiquinone oxidoreductase subunit B9) The protein encoded by this gene is a subunit of the mitochondrial oxidative phosphorylation complex I (nicotinamide adenine dinucleotide: ubiquinone oxidoreductase). Complex I is localized to the inner mitochondrial membrane and functions to dehydrogenate nicotinamide adenine dinucleotide and to shuttle electrons to coenzyme Q. Complex I deficiency is the most common defect found in oxidative phosphorylation disorders and results in a range of conditions, including lethal neonatal disease, hypertrophic cardiomyopathy, liver disease, and adult-onset neurodegenerative disorders. Pseudogenes of this gene are found on chromosomes five, seven and eight. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2015]

An alternative downstream in-frame start codon (Met44) is located in exon 2 of the encoded protein. An activation of potential downstream translation at this initiation site would result in a shortened protein missing the first 43 amino acids from the protein sequence. To our knowledge no pathogenic variants have been reported upstream of this alternate codon. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00014 in 251262 control chromosomes, predominantly at a frequency of 0.00029 within the Non-Finnish European subpopulation in the gnomAD database. This frequency does not allow for any conclusions about variant significance. To our knowledge, no occurrence of c.1A>G in individuals affected with Mitochondrial Complex 1 Deficiency, Nuclear Type 24 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 379528). Based on the evidence outlined above, the variant was classified as uncertain significance. -
