WRN (HGNC:12791): (WRN RecQ like helicase) This gene encodes a member of the RecQ subfamily of DNA helicase proteins. The encoded nuclear protein is important in the maintenance of genome stability and plays a role in DNA repair, replication, transcription and telomere maintenance. This protein contains a N-terminal 3' to 5' exonuclease domain, an ATP-dependent helicase domain and RQC (RecQ helicase conserved region) domain in its central region, and a C-terminal HRDC (helicase RNase D C-terminal) domain and nuclear localization signal. Defects in this gene are the cause of Werner syndrome, an autosomal recessive disorder characterized by accelerated aging and an elevated risk for certain cancers. [provided by RefSeq, Aug 2017]

The WRN p.Lys1087Glu variant was not identified in the literature nor was it identified in Cosmic or LOVD 3.0. The variant was identified in dbSNP (ID: rs374154973) and ClinVar (classified as uncertain significance by Invitae and Fulgent Genetics for Werner syndrome). The variant was identified in control databases in 54 of 279522 chromosomes (1 homozygous) at a frequency of 0.0001932 (Genome Aggregation Database March 6, 2019, v2.1.1). The variant was observed in the following populations: European (non-Finnish) in 47 of 128198 chromosomes (freq: 0.000367), African in 4 of 24796 chromosomes (freq: 0.000161), Latino in 2 of 34780 chromosomes (freq: 0.000058) and South Asian in 1 of 29552 chromosomes (freq: 0.000034), but was not observed in the Ashkenazi Jewish, East Asian, European (Finnish), or Other populations. The p.Lys1087 residue is not conserved in mammals and computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) do not suggest a high likelihood of impact to the protein; however, this information is not predictive enough to rule out pathogenicity.
