The c.2105C>T (p.A702V) alteration is located in exon 11 (coding exon 11) of the MCPH1 gene. This alteration results from a C to T substitution at nucleotide position 2105, causing the alanine (A) at amino acid position 702 to be replaced by a valine (V). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

The A702V variant has not been published as a pathogenic variant, nor has it been reported as a benign polymorphism to our knowledge. It was not observed in approximately 6,200 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The A702V substitution occurs at a position where amino acids with similar properties to Alanine are tolerated across species. However, the substitution is conservative, which is not likely to impact secondary protein structure as these residues share similar properties. In addition, missense variants at nearby residues have not been reported in the Human Gene Mutation Database in association with MCPH1-related disorders (Stenson et al., 2014). In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function.
