The encoded beta subunit appears to play a role in modulation of channel function in cone photoreceptors. This heterotetrameric channel is necessary for sensory transduction, and mutations in this gene have been associated with achromatopsia 3, progressive cone dystrophy, and juvenile macular degeneration, also known as Stargardt Disease. This sequence change replaces glycine, which is neutral and non-polar, with cysteine, which is neutral and slightly polar, at codon 558 of the CNGB3 protein (p.Gly558Cys). This variant is present in population databases (rs749413012, gnomAD 0.04%). This missense change has been observed in individual(s) with achromatopsia or retinitis pigmentosa (PMID: 15712225). ClinVar contains an entry for this variant (Variation ID: 554831). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on CNGB3 protein function. Experimental studies have shown that this missense change affects CNGB3 function (PMID: 26106334).
