This sequence change replaces valine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 663 of the NBN protein (p.Val663Leu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NBN-related conditions. ClinVar contains an entry for this variant (Variation ID: 461533). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.V663L variant (also known as c.1987G>C), located in coding exon 13 of the NBN gene, results from a G to C substitution at nucleotide position 1987. The valine at codon 663 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear. -
