The encoded protein is a member of the MRE11/RAD50 double-strand break repair complex which consists of 5 proteins. The p.P266R variant (also known as c.797C>G), located in coding exon 7 of the NBN gene, results from a C to G substitution at nucleotide position 797. The proline at codon 266 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
