This alteration results from a G to T substitution at nucleotide position 875, causing the arginine (R) at amino acid position 292 to be replaced by a leucine (L). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant was reported de novo in multiple individuals with clinical features consistent with STXBP1-related developmental and epileptic encephalopathy (Stamberger, 2016; Zaganas, 2021). Two alterations at the same codon, c.875G>A (p.R292H) and c.874C>T (p.R292C), have also been reported de novo in individuals with clinical features consistent with STXBP1-related developmental and epileptic encephalopathy (Michaud, 2014; Stamberger, 2016; Uddin, 2017). This amino acid position is highly conserved in available vertebrate species. This missense alteration is located in a region that has a low rate of benign missense variation (Lek, 2016; Firth, 2009). This alteration is predicted to be deleterious by in silico analysis.
