The proline at codon 1553 is replaced by leucine, an amino acid with some similar properties. Based on data from gnomAD, the T allele has an overall frequency of approximately 0.01% (36/277222). The highest observed frequency was 0.16% (16/10152) of Ashkenazi Jewish alleles. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
