EHMT1 (HGNC:24650): (euchromatic histone lysine methyltransferase 1) The protein encoded by this gene is a histone methyltransferase that methylates the lysine-9 position of histone H3. This action marks the genomic region packaged with these methylated histones for transcriptional repression. This protein may be involved in the silencing of MYC- and E2F-responsive genes and therefore could play a role in the G0/G1 cell cycle transition. Defects in this gene are a cause of chromosome 9q subtelomeric deletion syndrome (9q-syndrome, also known as Kleefstra syndrome). Alternative splicing results in multiple transcript variants. The c.871C>T (p.R291*) alteration, located in exon 5 (coding exon 5) of the EHMT1 gene, consists of a C to T substitution at nucleotide position 871. This changes the amino acid from an arginine (R) to a stop codon at amino acid position 291. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant was reported in individual(s) with features consistent with EHMT1-related Kleefstra syndrome; in at least one individual, it was determined to be de novo (Kleefstra, 2009; Rots, 2024). Based on the available evidence, this alteration is classified as pathogenic. -
