This RNA also interacts with the telomerase reverse transcriptase catalytic subunit to form a distinct ribonucleoprotein complex that has RNA-dependent RNA polymerase activity and produces double-stranded RNAs that can be processed into small interfering RNA. This variant has been observed in individual(s) with clincial features of cartilage-hair hypoplasia anauxetic dysplasia (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 465203). This variant disrupts the n.181G nucleotide in the RMRP gene. Other variant(s) that disrupt this nucleotide have been determined to be pathogenic (PMID: 16254002, 16838329, 27862957). This suggests that this nucleotide is clinically significant, and that variants that disrupt this position are likely to be disease-causing.
