Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251382 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2324A>G has been reported in the literature in an individuals affected with Glycine Encephalopathy (Non-Ketotic Hyperglycinemia; Conter_2006, Coughlin_2017). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. However, a different variant at the same amino acid has been reported in association with Hyperglycinaemia, non-ketotic (p.H775P, HGMD).
