Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 1.2e-05 in 251382 control chromosomes. c.2324A>G has been observed in individual(s) affected with Glycine Encephalopathy (Non-Ketotic Hyperglycinemia; Conter_2006, Coughlin_2017). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. However, variants affecting the same amino acid (p.H775P, p.H775Y) have been observed in individuals with Non-Ketotic Hyperglycinemia (HGMD, internal data), suggesting this residue may be important for GLDC function.
