Variant summary: FXN c.460A>T (p.Ile154Phe) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251460 control chromosomes. c.460A>T has been reported in the literature in individuals affected with Friedreich Ataxia. These data indicate that the variant is likely to be associated with disease (Filla_AJHG, Sacca_2011). At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in significantly reduced allosteric activation in binding and activating the SDU complex via in vitro Binding/Biosynthesis assays (Tsai_2011). The following publications have been ascertained in the context of this evaluation (PMID: 8751856, 21412413, 21671584). ClinVar contains an entry for this variant (Variation ID: 3981).
