The threonine at codon 515 is replaced by serine, an amino acid with similar properties. In one study, this alteration was detected in 1/1441 breast cancer cases and 0/464 healthy controls (Thompson ER et al. PLoS Genet, 2012 Sep;8:e1002894). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
