The p.L486S variant (also known as c.1457T>C), located in coding exon 9 of the TGFBR1 gene, results from a T to C substitution at nucleotide position 1457. The leucine at codon 486 is replaced by serine, an amino acid with dissimilar properties. This variant was detected in on individual with a type A dissection and was found in 4 additional relatives with ascending aortic aneurysms (Tran-Fadulu V et al. J. Med. Genet., 2009 Sep;46:607-13). This amino acid position is highly conserved in available vertebrate species. This variant was not reported in the gnomAD database, with coverage at this position. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic. -

This sequence change replaces leucine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 486 of the TGFBR1 protein (p.Leu486Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with thoracic aortic aneurysm and dissection (PMID: 19542084, 30675029).
