The p.Ala577Val var iant in TMPO has not been previously reported in individuals with cardiomyopathy , but has been identified in 0.2% (24/10554) of African chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs34150443) . Computational prediction tools and conservation analysis suggest that this var iant may impact the protein, though this information is not predictive enough to determine pathogenicity. In summary, while the clinical significance of the p.A la577Val variant is uncertain, its frequency suggests that it is more likely to be benign.
The p.A577V variant (also known as c.1730C>T), located in coding exon 4 of the TMPO gene, results from a C to T substitution at nucleotide position 1730. The alanine at codon 577 is replaced by valine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
