It heteromultimerizes with another MAGUK protein, DLG2, and is recruited into NMDA receptor and potassium channel clusters. These two MAGUK proteins may interact at postsynaptic sites to form a multimeric scaffold for the clustering of receptors, ion channels, and associated signaling proteins. This variant has been observed in individual(s) with clinical features of DLG4-related conditions (Invitae). In at least one individual the variant was observed to be de novo. For these reasons, this variant has been classified as Pathogenic. Donor and acceptor splice site variants typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in DLG4 are known to be pathogenic (PMID: 27479843, 29460436). Experimental studies and prediction algorithms are not available or were not evaluated, and the effect of this variant on mRNA splicing is currently unknown. This variant is not present in population databases (ExAC no frequency). This sequence change affects an acceptor splice site in intron 20 of the DLG4 gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. -
