The p.Glu15Gln variant in USH1G has not been previously reported in individuals with hearing loss. This variant has been identified in 1/16714 European chromos omes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org). Although this variant has been seen in the general population, its frequency is not high enough to rule out a pathogenic role. Computational prediction tools a nd conservation analyses do not provide strong support for or against an impact to the protein. In summary, the clinical significance of the p.Glu15Gln variant is uncertain. -

This sequence change replaces glutamic acid with glutamine at codon 15 of the USH1G protein (p.Glu15Gln). The glutamic acid residue is moderately conserved and there is a small physicochemical difference between glutamic acid and glutamine. This variant is present in population databases (rs757653096, ExAC 0.006%). This variant has not been reported in the literature in individuals with USH1G-related conditions. ClinVar contains an entry for this variant (Variation ID: 229608). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
