Inborn genetic diseases Uncertain:1
The p.M358T variant (also known as c.1073T>C), located in coding exon 5 of the RECQL4 gene, results from a T to C substitution at nucleotide position 1073. The methionine at codon 358 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
Baller-Gerold syndrome Uncertain:1
This sequence change replaces methionine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 358 of the RECQL4 protein (p.Met358Thr). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt RECQL4 protein function. ClinVar contains an entry for this variant (Variation ID: 568336). This variant has not been reported in the literature in individuals affected with RECQL4-related conditions. This variant is present in population databases (rs376652305, gnomAD 0.003%). -
