The PSEN1 c.691G>T variant is predicted to result in the amino acid substitution p.Ala231Ser. To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. Other missense changes at the same amino acid position, c.691G>A (Ala231Thr) and c.692C>T (Ala231Val) have previously been reported to be causative for Alzheimer's Disease (Campion et al 1995. PubMed ID: 8634712; Cruts et al 1998. PubMed ID: 9384602). Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
