The p.S1631C variant (also known as c.4891A>T), located in coding exon 15 of the APC gene, results from an A to T substitution at nucleotide position 4891. The serine at codon 1631 is replaced by cysteine, an amino acid with dissimilar properties. This alteration has been identified in an individual with polyposis (Marabelli M et al. Genet Test Mol Biomarkers, 2016 12;20:777-785). This amino acid position is not well conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces serine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 1631 of the APC protein (p.Ser1631Cys).
