This missense change has been observed in individuals with clinical features of autosomal dominant autoimmune lymphoproliferative syndrome (PMID: 9927496; Invitae). This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 260 of the FAS protein (p.Asp260Gly). This variant is not present in population databases (gnomAD no frequency). This variant is also known as D244G. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Not Available"). Experimental studies have shown that this missense change affects FAS function (PMID: 9927496). This variant disrupts the p.Asp260 amino acid residue in FAS. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 9821419, 18223337, 20935634, 21490157; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively.
