The arylsulfatase B homodimer hydrolyzes sulfate groups of N-Acetyl-D-galactosamine, chondriotin sulfate, and dermatan sulfate. The protein is targeted to the lysozyme. Mucopolysaccharidosis type VI is an autosomal recessive lysosomal storage disorder resulting from a deficiency of arylsulfatase B. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Donor and acceptor splice site variants typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in ARSB are known to be pathogenic (PMID: 17458871, 22133300). Disruption of this splice site has been observed in an individual affected with mucopolysaccharidosis type VI (PMID: 17161971). This variant is not present in population databases (ExAC no frequency). This sequence change affects an acceptor splice site in intron 6 of the ARSB gene. It is expected to disrupt RNA splicing and likely results in an absent or disrupted protein product. -
