Variant summary: ASL c.209T>C (p.Val70Ala) results in a non-conservative amino acid change located in the fumarate lyase, N-terminal domain (IPR022761) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. Several computational tools predict a significant impact on normal splicing: Three predict the variant weakens a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 244992 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.209T>C has been reported in the literature in at least an individual affected with argininosuccinic aciduria (example: Balmer_2014). These report(s) do not provide unequivocal conclusions about association of the variant with Argininosuccinic Aciduria. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
