The p.R270K variant (also known as c.809G>A), located in coding exon 6 of the ATM gene, results from a G to A substitution at nucleotide position 809. The arginine at codon 270 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces arginine, which is basic and polar, with lysine, which is basic and polar, at codon 270 of the ATM protein (p.Arg270Lys). This variant is present in population databases (rs587781857, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with ATM-related conditions. ClinVar contains an entry for this variant (Variation ID: 141582).
