This variant has been reported in an individual affected with pancreatic cancer (PMID: 25940717). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
The p.L579* pathogenic mutation (also known as c.1736T>G), located in coding exon 9 of the BRCA2 gene, results from a T to G substitution at nucleotide position 1736. This changes the amino acid from a leucine to a stop codon within coding exon 9. This alteration has been reported as a germline mutation in multiple individuals with pancreatic cancer (Golan T et al. Br. J. Cancer, 2014 Sep;111:1132-8; Lohse I et al. Br. J. Cancer, 2015 Jul;113:425-32; Holter S et al. J. Clin. Oncol., 2015 Oct;33:3124-9). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
