The BRCA2 c.2464T>C (p.Cys822Arg) variant has been reported in families with a high risk of hereditary breast and/or ovarian cancer PMID: 21147080 (2011), 28477318 (2017)). In a large breast cancer association study, this variant was found in an individual affected with breast cancer (PMID: 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared)). It has also been described to be located in a region of the BRCA2 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). The frequency of this variant in the general population, 0.0000041 (1/241802 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant.
