This missense variant replaces valine with isoleucine at codon 1186 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with ovarian cancer (PMID: 34157791). A multifactorial analysis has reported a likelihood ratio for pathogenicity based on personal and family history of 0.886 (based on log(LR) = -0.052446544) (PMID: 31853058). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
