The c.5984dupA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a duplication of A at nucleotide position 5984, causing a translational frameshift with a predicted alternate stop codon (p.N1995KFS*8). This mutation has been reported in Iranian breast cancer families (Keshavarzi F et al, Fam. Cancer 2012 Mar; 11(1):57-67). In addition to the clinical data presented in the literature, since premature stop codons are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294).
Hereditary breast ovarian cancer syndrome Pathogenic:1
For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 51981). This premature translational stop signal has been observed in individual(s) with hereditary breast and/or ovarian cancer (PMID: 21918854, 24549055). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Asn1995Lysfs*8) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584).
