The BRCA2, p.Leu2165Leu variant was not identified in the literature, nor was it identified in the dbSNP, NHLBI Exome Sequencing Project (Exome Variant Server), Exome Aggregation Consortium (ExAC), Fanconi Anaemia LOVD, COSMIC, ClinVar, Clinvitae, GeneInsight VariantWire through the Canadian Open Genetics Repository (http://opengenetics.ca/), BIC, ARUP Laboratories, MutDB or BRCA Share UMD databases. The p.Leu2165Leu variant is not expected to have clinical significance because it does not result in a change of amino acid and is not located in a known consensus splice site. One of five in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer, HumanSpliceFinder) predict the loss of a cryptic 5’ donor site and one of five of the above programs predict the creation of a cryptic 3’ acceptor site.
