Variant summary: BRCA2 c.8331+16C>G alters a non-conserved nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 2.9e-05 in 245230 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.c.8331+16C>G has been reported in the literature in individuals affected with Hereditary Breast and Ovarian Cancer. These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast and Ovarian Cancer (example, Levene_2003, Hansen_2011, Thery_2011,). At-least two co-occurrences with pathogenic variants have been reported in the UMD database and in the literature [BRCA2 c.7008-1G>A (UMD database); BRCA1 c.68_69delAG, p.Glu23ValfsX17 (Hansen_2011)], providing supporting evidence for a benign role. The following publications have been ascertained in the context of this evaluation (PMID: 21318380, 14574163, 21673748).ClinVar contains an entry for this variant (Variation ID: 182298). Based on the evidence outlined above, the variant was classified as likely benign.
