DNA sequence analysis of the BRCA2 gene demonstrated a sequence change in intron 18, c.8332-9T>G. This change does not appear to have been previously described in individuals with BRCA2-related disorders and has also not been described in population databases such as ExAC and gnomAD. In-silico splice prediction programs provide inconclusive results for this sequence change. It is possible that this sequence change represents a benign sequence change in the BRCA2 gene that has not been identified to date. The functional significance of this sequence change is not known at present and its contribution to this individual's disease phenotype cannot definitively be determined.
The BRCA2 c.8332-9T>G variant was not identified in the literature nor was it identified in the dbSNP, Cosmic, LOVD 3.0, UMD-LSDB, BIC Database, ARUP Laboratories, or Zhejiang University, databases. The variant was identified in ClinVar (classified as uncertain significance by COGR) database. The variant was not identified in the following control databases: the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The c.8332-9T>G variant is located in the 3' splice region but does not affect the invariant -1 and -2 positions. However, positions -3 and -5 to -12 are part of the splicing consensus sequence and variants involving these positions sometimes affect splicing.
