Hereditary cancer-predisposing syndrome Uncertain:1
The p.F32L variant (also known as c.94T>C) is located in coding exon 2 of the BRCA2 gene. This alteration results from a T to C substitution at nucleotide position 94. The phenylalanine at codon 32 is replaced by leucine, an amino acid with highly similar properties. This alteration was identified in a Japanese family with cases of both female and male breast cancers, however it's clinical significance was unclear (Katagiri, T et al. J Hum Genet. 1998;43(1):42-8). Based on protein sequence alignment, this amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. However, since supporting evidence is limited at this time, the clinical significance of p.F32L remains unclear.​ -

In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function. ClinVar contains an entry for this variant (Variation ID: 52851). This missense change has been observed in individual(s) with breast cancer (PMID: 9609997, 25503501). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 32 of the BRCA2 protein (p.Phe32Leu). -
