This gene encodes a muscle-specific member of the calpain large subunit family that specifically binds to titin. Mutations in this gene are associated with limb-girdle muscular dystrophies type 2A. The c.371G>C (p.G124A) alteration is located in exon 2 (coding exon 2) of the CAPN3 gene. This alteration results from a G to C substitution at nucleotide position 371, causing the glycine (G) at amino acid position 124 to be replaced by an alanine (A). for autosomal recessive limb-girdle muscular dystrophy; however, its clinical significance for autosomal dominant limb-girdle muscular dystrophy is uncertain. Based on data from gnomAD, the C allele has an overall frequency of 0.001% (3/251394) total alleles studied. This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, this alteration is classified as likely pathogenic.
