Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 9361025, 9889017, 10338090, 14722927). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Experimental studies have shown that this missense change affects CBS function (PMID: 20506325, 25331909). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CBS protein function. This missense change has been observed in individual(s) with homocystinuria (PMID: 11013450). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces threonine, which is neutral and polar, with arginine, which is basic and polar, at codon 262 of the CBS protein (p.Thr262Arg). -
