NM_000090.4:c.3575_3576insAGGG

Variant summary

Our verdict is Pathogenic. The variant received 12 ACMG points: 12P and 0B. PVS1PM2PP5_Moderate

The NM_000090.4(COL3A1):c.3575_3576insAGGG(p.Pro1193GlyfsTer20) variant causes a frameshift change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. Variant has been reported in ClinVar as Pathogenic (★). Synonymous variant affecting the same amino acid position (i.e. A1192A) has been classified as Likely benign. Variant results in nonsense mediated mRNA decay.

Genes affected
COL3A1 (HGNC:2201): (collagen type III alpha 1 chain) This gene encodes the pro-alpha1 chains of type III collagen, a fibrillar collagen that is found in extensible connective tissues such as skin, lung, uterus, intestine and the vascular system, frequently in association with type I collagen. Mutations in this gene are associated with Ehlers-Danlos syndrome type IV, and with aortic and arterial aneurysms. [provided by R. Dalgleish, Feb 2008]

The c.3575_3576insAGGG pathogenic mutation, located in coding exon 48 of the COL3A1 gene, results from an insertion of 4 nucleotides at position 3575, causing a translational frameshift with a predicted alternate stop codon (p.P1193Gfs*20). This variant was reported in individual(s) with features consistent with vascular Ehlers-Danlos syndrome (Leistritz DF et al. Genet Med, 2011 Aug;13:717-22). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
