The p.P1833S variant (also known as c.5497C>T), located in coding exon 66 of the COL5A1 gene, results from a C to T substitution at nucleotide position 5497. The proline at codon 1833 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.

This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 1833 of the COL5A1 protein (p.Pro1833Ser). This variant is present in population databases (rs370789226, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with COL5A1-related conditions. ClinVar contains an entry for this variant (Variation ID: 459716).
