For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this missense change affects CYP17A1 function (PMID: 14715827). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 1797). This missense change has been observed in individual(s) with 17-alpha-hydroxylase deficiency (PMID: 14671162, 14715827). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces tryptophan, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 406 of the CYP17A1 protein (p.Trp406Arg). -
