Therefore, this variant has been classified as Likely Pathogenic. This variant disrupts the p.Tyr969 amino acid residue in MSH6. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 20587412, 16813607, 19723918, 23621914). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. This variant has not been reported in the literature in individuals with MSH6-related conditions. ClinVar contains an entry for this variant (Variation ID: 433916). This variant is not present in population databases (ExAC no frequency). This sequence change replaces tyrosine with histidine at codon 969 of the MSH6 protein (p.Tyr969His). The tyrosine residue is highly conserved and there is a moderate physicochemical difference between tyrosine and histidine.
