ClinVar reports functional evidence for this variant: "SCV000280144: cells transfected with this mutant protein showed no delayed rectifying potassium current. Immunoblots of cell lysates showed that the KCNQ1 Tyr461Stop subunit cannot form channel tetramers by itself or with the WT subunit. Also, immunocytochemical analysis showed that the surface expression of this mutant subunit was very low with or without WT subunit present. They conclude that loss of the C-terminal domain of the protein impairs the assembly, trafficking, and function of the mutant subunit-containing channels but does not interfere with expression of the WT channel. Kimoto K et al. (2013)". Synonymous variant affecting the same amino acid position (i.e. Y461Y) has been classified as Likely benign. Variant results in nonsense mediated mRNA decay.
