The Ser581Stop mutation in the KCNH2 gene has been reported previously in one individual diagnosed with LQTS and absent from 2,600 control alleles. This mutation is predicted to cause loss of normal protein function either due to production of an abnormal, prematurely truncated protein, or by absence of protein product due to nonsense mediated mRNA decay. Multiple nonsense mutations in the KCNH2 gene have been reported previously in association with LQTS. The variant is found in LQT panel(s). -
Long QT syndrome Pathogenic:1
This sequence change creates a premature translational stop signal (p.Ser581*) in the KCNH2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in KCNH2 are known to be pathogenic (PMID: 10973849, 19862833). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with clinical features of long QT syndrome (PMID: 19716085). ClinVar contains an entry for this variant (Variation ID: 200737). For these reasons, this variant has been classified as Pathogenic. -
