NM_000245.4:c.1975A>G
Variant summary
Our verdict is Uncertain significance. The variant received 1 ACMG points: 2P and 1B. PM2BP4
The NM_000245.4(MET):c.1975A>G(p.Ile659Val) variant causes a missense change. The variant allele was found at a frequency of 0.000000684 in 1,461,720 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. I659L) has been classified as Likely benign.

The p.I659V variant (also known as c.1975A>G), located in coding exon 7 of the MET gene, results from an A to G substitution at nucleotide position 1975. The isoleucine at codon 659 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
