The p.Q562P variant (also known as c.1685A>C), located in coding exon 15 of the MLH1 gene, results from an A to C substitution at nucleotide position 1685. The glutamine at codon 562 is replaced by proline, an amino acid with similar properties. This variant has been reported in 1 individual in an Lynch Syndrome cohort, but clinical details were limited (Roht L et al. Cancers (Basel), 2023 Jul;15:). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
Hereditary nonpolyposis colorectal neoplasms Pathogenic:1
This sequence change replaces glutamine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 562 of the MLH1 protein (p.Gln562Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of Lynch syndrome (PMID: 21404117; internal data). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 547537). Invitae Evidence Modeling incorporating data from in vitro experimental studies (internal data) indicates that this missense variant is expected to disrupt MLH1 function with a positive predictive value of 95%.
