Genes affected
MSH2 (HGNC:7325): (mutS homolog 2) This locus is frequently mutated in hereditary nonpolyposis colon cancer (HNPCC). When cloned, it was discovered to be a human homolog of the E. coli mismatch repair gene mutS, consistent with the characteristic alterations in microsatellite sequences (RER+ phenotype) found in HNPCC. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Apr 2012]

This sequence change creates a premature translational stop signal (p.Cys176Profs*2) in the MSH2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MSH2 are known to be pathogenic (PMID: 15849733, 24362816). For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 428549). This variant has not been reported in the literature in individuals affected with MSH2-related conditions. This variant is not present in population databases (gnomAD no frequency).

Hereditary cancer-predisposing syndrome Pathogenic:1

Significance:Pathogenic
Review Status:criteria provided, single submitter
Collection Method:clinical testing

The c.525_532delGTGTGAAT pathogenic mutation, located in coding exon 3 of the MSH2 gene, results from a deletion of 8 nucleotides at nucleotide positions 525 to 532, causing a translational frameshift with a predicted alternate stop codon (p.C176Pfs*2). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
