Hypertrophic cardiomyopathy Pathogenic:1
The c.2994+2T>C variant in MYBPC3 has been reported in one individual with hypertrophic cardiomyopathy (HCM) and segregated with disease in at least one affected relative (Ho 2018 PMID: 30297972, LMM data). It was absent from large population studies. This variant occurs within the canonical splice site (+/- 1,2) and is predicted to cause altered splicing leading to an abnormal or absent protein. Loss of function of the MYBPC3 gene is an established disease mechanism in autosomal dominant HCM. In summary, although additional studies are required to fully establish its clinical significance, this variant meets criteria to be classified as likely pathogenic for autosomal dominant HCM.
