Variant summary: PTCH1 c.2834G>A (p.Arg945Gln) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant allele was found at a frequency of 1.5e-05 in 1614020 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in PTCH1 causing Nevoid Basal Cell Carcinoma Syndrome (Gorlin Syndrome) (1.5e-05 vs 1.7e-05), allowing no conclusion about variant significance. c.2834G>A has been observed in at least one individual affected with breast cancer (Zhunussova_2023). These report(s) do not provide unequivocal conclusions about association of the variant with Nevoid Basal Cell Carcinoma Syndrome (Gorlin Syndrome). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
