The p.R1025H variant (also known as c.3074G>A), located in coding exon 18 of the PTCH1 gene, results from a G to A substitution at nucleotide position 3074. The arginine at codon 1025 is replaced by histidine, an amino acid with highly similar properties. This alteration was detected in a pediatric patient with myelodysplastic syndrome (MDS) (Schwartz JR et al. Nat Commun, 2017 11;8:1557). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
