The p.I1148L variant (also known as c.3442A>C), located in coding exon 20 of the PTCH1 gene, results from an A to C substitution at nucleotide position 3442. The isoleucine at codon 1148 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear. -
