ClinVar contains an entry for this variant (Variation ID: 550833). This missense change has been observed in individual(s) with Niemann-Pick disease type C (PMID: 26981555). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 1045 of the NPC1 protein (p.Leu1045Pro). -

Variant summary: NPC1 c.3134T>C (p.Leu1045Pro) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251442 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3134T>C has been reported in the literature in at least one individual affected with Niemann-Pick Disease Type C. These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
