The encoded protein is believed to be an integral membrane protein involved in small molecule transport, specifically tyrosine, which is a precursor to melanin synthesis. It is involved in mammalian pigmentation, where it may control skin color variation and act as a determinant of brown or blue eye color. This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 179 of the OCA2 protein (p.Lys179Glu). This variant is present in population databases (no rsID available, gnomAD 0.01%). This missense change has been observed in individuals with albinism (PMID: 25119903, 34838614). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 436096). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt OCA2 protein function with a positive predictive value of 80%. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively.
